  1. Before running the reaction it is important to barcode both sequencing plates and put them onto LIMS. If you forget to do this before thermocycling, you can do it after you ran the reaction, though that is not ideal because the fluorescent markers degrade quickly when exposed to light.
  2. Go to LIMS, and under the section Stage Selection select Cycle Sequencing Prep. User barcode: Scan in your card. Start plate: scan the PCR plate. End plate: scan the sequencing plate. Put the barcode on underneath the very edge of the plate. Select the protocol. Submit. You have to do this twice, once for forward and once for reverse.
  3. Go to LIMS again, and under Stage Selection select Cycle Sequence Thermocycler. User Barcode: Scan your card. Plate Barcode: scan the sequencing plate. Protocol: SEQ31 standard CS thermocycling (the only option). Submit. You have to do this twice, once for forward and once for reverse.
